Biopharmaceutical investigation of Diclofenac sodium suppositories, prepared on varied bases by different methods, including suppositories of industrial production, was carried out. Essential dependence of Diclofenac sodium release from suppositories (Dissolution test) on a base composition was established. Cocoa butter: hydrogenated palm oil base (1:1) and Witepsol have demonstrated the highest completeness and rate of release among oily bases to be investigated. Introduction into hard fat of such surface – active substances as emulsifying agents T-2, N 1, phospholipid concentrate allowed to achieve the release from suppositories up to 100%. From the biopharmaceutical point of view the mentioned above excipients were optimal for the Diclofenac sodium suppositories manufacturing and they could be alternative to Witepsol. The dependence between the softening time and the rate and release completeness of the ortofen from the suppositories was not established.

Key words: suppositories, Diclofenac sodium, Dissolution test, Witepsol, Cocoa butter, Hard fat, surface-active agents
